7Download%27%20class%3D%27zp-DownloadURL%27%20href%3D%27https%3A%5C%2F%5C%2Fwww.welchbiology.com%5C%2Fwp-content%5C%2Fplugins%5C%2Fzotpress%5C%2Flib%5C%2Frequest%5C%2Frequest.dl.php%3Fapi_user_id%3D5126420%26amp%3Bdlkey%3DX4DN9A5Y%26amp%3Bcontent_type%3Dapplication%5C%2Fpdf%27%3EDownload%3C%5C%2Fa%3E%3C%5C%2Fdiv%3E%5Cn%3C%5C%2Fdiv%3E%22%2C%22data%22%3A%7B%22itemType%22%3A%22journalArticle%22%2C%22title%22%3A%22Critical%20oxygen%20tensions%20as%20predictors%20of%20hypoxia%20tolerance%20and%20tissue%20metabolic%20responses%20during%20hypoxia%20exposure%20in%20fishes%22%2C%22creators%22%3A%5B%7B%22creatorType%22%3A%22author%22%2C%22firstName%22%3A%22Ben%22%2C%22lastName%22%3A%22Speers-Roesch%22%7D%2C%7B%22creatorType%22%3A%22author%22%2C%22firstName%22%3A%22Milica%22%2C%22lastName%22%3A%22Mandic%22%7D%2C%7B%22creatorType%22%3A%22author%22%2C%22firstName%22%3A%22Derrick%20J.%20E.%22%2C%22lastName%22%3A%22Groom%22%7D%2C%7B%22creatorType%22%3A%22author%22%2C%22firstName%22%3A%22Jeffrey%20G.%22%2C%22lastName%22%3A%22Richards%22%7D%5D%2C%22abstractNote%22%3A%22An%20organism%27s%20critical%20oxygen%20tension%20%28Pcrit%29%20reflects%20its%20ability%20to%20extract%20environmental%20O2.%20Consequently%2C%20Pcrit%20has%20been%20used%20as%20an%20indicator%20of%20hypoxia%20tolerance%20in%20aquatic%20animals.%20The%20relationship%20between%20Pcrit%20and%20hypoxia%20tolerance%20and%20hypoxic%20metabolic%20responses%2C%20however%2C%20remains%20incompletely%20understood.%20Among%20several%20species%20of%20sculpin%20fishes%20%28superfamily%20Cottoidea%29%2C%20we%20previously%20demonstrated%20a%20correlation%20between%20Pcrit%20and%20hypoxia%20tolerance%2C%20as%20measured%20as%20the%20time%20required%20for%2050%25%20of%20a%20group%20of%20fish%20to%20lose%20equilibrium%20%28LOE50%29%20at%20a%20water%20PO2%20of%206.4Torr.%20In%20the%20present%20study%2C%20we%20further%20investigated%20the%20relationship%20between%20Pcrit%2C%20hypoxia%20tolerance%2C%20and%20hypoxic%20metabolic%20responses%20by%20examining%20the%20effects%20of%20hypoxia%20exposure%20at%20a%20fixed%20percentage%20of%20Pcrit%20%2830%25%3B%20termed%20relative%20hypoxia%20exposure%29%20on%20LOE50%20and%20metabolic%20responses%20in%20brain%2C%20liver%2C%20and%20white%20muscle%20in%20three%20sculpin%20species%20that%20differ%20in%20Pcrit%20and%20hypoxia%20tolerance%20at%206.4Torr.%20We%20also%20assessed%20the%20tissue%20metabolic%20responses%20underlying%20hypoxic%20loss%20of%20equilibrium%20%28LOE%29.%20The%20species%2C%20from%20most%20to%20least%20hypoxia-tolerant%20at%206.4Torr%20and%20from%20lowest%20to%20highest%20Pcrit%20values%2C%20were%20the%20tidepool%20sculpin%20%28Oligocottus%20maculosus%29%2C%20staghorn%20sculpin%20%28Leptocottus%20armatus%29%2C%20and%20silverspotted%20sculpin%20%28Blepsias%20cirrhosus%29.%20If%20Pcrit%20predicts%20hypoxia%20tolerance%2C%20then%20we%20expected%20similar%20LOE50%20values%20and%20similar%20tissue%20metabolic%20responses%20across%20all%20species%20during%20relative%20hypoxia%20exposure.%20LOE50%20values%20were%20similar%20in%20staghorn%20sculpins%20and%20tidepool%20sculpins%2C%20but%20not%20in%20silverspotted%20sculpins%2C%20which%20had%20a%20comparatively%20lower%20relative%20hypoxia%20LOE50%20value.%20Thus%2C%20Pcrit%2C%20and%20consequently%20the%20ability%20to%20extract%20environmental%20O2%2C%20cannot%20predict%20hypoxia%20tolerance%20in%20all%20species%2C%20at%20least%20at%20a%20water%20PO2%20of%2030%25%20of%20Pcrit.%20During%20relative%20hypoxia%20exposure%2C%20tissue%20lactate%20accumulation%20and%20ATP%20levels%20were%20similar%20between%20species%2C%20suggesting%20that%20the%20ability%20to%20extract%20environmental%20O2%20is%20an%20important%20determinant%20of%20cellular%20energy%20status%20and%20reliance%20on%20anaerobic%20glycolysis%20in%20hypoxic%20sculpins.%20However%2C%20whereas%20tissue%20glycogen%20content%20and%20utilization%20were%20similar%20between%20tidepool%20sculpins%20and%20staghorn%20sculpins%2C%20there%20were%20lower%20normoxic%20levels%20and%20greater%20hypoxic%20depletion%20in%20silverspotted%20sculpins%2C%20potentially%20explaining%20their%20poorer%20relative%20hypoxia%20tolerance.%20In%20all%20species%2C%20LOE%20was%20associated%20with%20depletion%20of%20brain%20%5BATP%5D.%20Overall%2C%20caution%20is%20warranted%20when%20Pcrit%20is%20used%20as%20an%20indicator%20of%20hypoxia%20tolerance%2C%20especially%20when%20considering%20temporal%20aspects%20of%20hypoxia%20tolerance%20and%20related%20metabolic%20characteristics%20%28e.g.%20glycogen%20availability%29.%20Ideally%2C%20comparative%20studies%20of%20hypoxia%20tolerance%20should%20feature%20multiple%20measures%20%28e.g.%20Pcrit%20and%20LOE50%29%20in%20order%20to%20assess%20the%20overall%20responses%20of%20fishes%20to%20hypoxia.%22%2C%22date%22%3A%22November%201%2C%202013%22%2C%22language%22%3A%22%22%2C%22DOI%22%3A%2210.1016%5C%2Fj.jembe.2013.10.006%22%2C%22ISSN%22%3A%220022-0981%22%2C%22url%22%3A%22http%3A%5C%2F%5C%2Fwww.sciencedirect.com%5C%2Fscience
